Usage and dosage

▸ Intramuscular or subcutaneous injection: Single dose: Horses 4-10ml; Cattle 4-20ml; Sheep and Pigs 2-5ml; Dogs 0.25-1ml.

Drug Interactions

▸This product can prolong and enhance the muscle relaxation effect of depolarizing succinylcholine chloride.

▸It has an antagonistic effect with non-depolarizing muscle relaxants.

Pharmacological effects

Neostigmine inhibits cholinesterase activity, preventing the hydrolysis of acetylcholine and increasing its concentration in the body, thereby enhancing and prolonging its effect. Neostigmine has a strong stimulating effect on the smooth muscles of the gastrointestinal tract, bladder, and uterus, but a weaker stimulating effect on glandular, iris, and bronchial smooth muscles, and a weaker inhibitory effect on the cardiovascular system; its effect on the central nervous system is not significant. Neostigmine has the strongest effect on skeletal muscle. This skeletal muscle stimulation is related not only to its inhibition of cholinesterase but also to its direct stimulation of N2 receptors on the motor endplates and its promotion of acetylcholine release from motor nerve endings. Neostigmine is poorly absorbed orally and does not easily cross the blood-brain barrier. Its plasma protein binding rate is 15%–25%. In the body, some of the drug is hydrolyzed by plasma cholinesterase and excreted in the urine as quaternary ammonium alcohols and unchanged drug. The portion metabolized by the liver is excreted through the bile duct.

Adverse reactions

Therapeutic doses have few side effects. Overdose can cause sweating, bradycardia, muscle tremors, or muscle paralysis.

Functions and uses

Cholinergic drugs. Primarily used for gastrointestinal atony, myasthenia gravis, and retained placenta.

Precautions

▸This product is contraindicated in animals with mechanical intestinal obstruction or bronchial asthma.

▸Atropine can be used to counteract its excitatory effect on M receptors in cases of poisoning.
