Pharmacological Action

After the progesterone vaginal release insert is placed into the vagina, progesterone is gradually released and absorbed through the vaginal mucosa into the body. This maintains plasma progesterone concentrations at normal effective levels and produces the same effect as endogenous progesterone. Through a feedback mechanism, it suppresses the secretion of pituitary gonadotropins and hypothalamic gonadotropin-releasing hormone (GnRH), thereby inhibiting estrus and ovulation and artificially prolonging the luteal phase. Once the progesterone release insert is removed, the effect of progesterone disappears, the pituitary begins to secrete gonadotropins, and follicular growth and estrus are promoted. This product may be used in combination with estrogens, gonadotropin-releasing hormone (GnRH), and prostaglandins.
Indications and Usage
For the control of the estrous cycle in nulliparous and parous heifers/cows. Indicated for estrus synchronizing and embryo transfer in cattle, as well as treatment of anestrus postpartum and during lactation.
Dosage and Administration
Route: Intravaginal use
▸ Cattle: One insert per animal. Remove after 5–8 days.
Placement Method
1. Rinse the vaginal applicator with a non-irritating disinfectant solution, then immerse the tip of the applicator in veterinary obstetrical lubricant.
2. Load the progesterone insert into the applicator. The wings of the insert should be closed, with only the wing tips protruding slightly from the applicator tip.
3. Immerse the tip of the loaded applicator in veterinary obstetrical lubricant.
4. Lift the animal's tail and clean the vulva with a disposable tissue.
5. Insert the applicator tip slightly upwards into the vulva and gently advance it into the anterior vagina.
6. Firmly squeeze the applicator handle to release the insert, then withdraw the applicator.
7. When correctly positioned, the wings of the insert will open in the anterior vagina, and the tail of the insert should remain visible outside the vulva.
Removal Method
Grasp the tail of the insert and pull gently and steadily to remove it. Used inserts should be incinerated or deeply buried.
For Synchronizing Estrus in Various Cows
▸ Day 0: Insert the progesterone insert. Gonadotropin-releasing hormone (GnRH) may be injected concurrently.
▸ Day 8: Remove the progesterone insert and inject prostaglandin simultaneously (nulliparous heifers require only half the dose).
▸ Day 9: Inject GnRH.
Most animals will begin estrus on Day 10. Fixed-time artificial insemination should be performed 12–20 hours after GnRH injection on Day 10 for all dairy heifers, beef heifers, and beef cows. For lactating dairy cows, artificial insemination is recommended upon observed estrus between Days 10–12.
For Treating Anovulatory Anestrus in Lactating Dairy Cows
▸ Day 0: Insert the progesterone insert.
▸ Day 6: Remove the progesterone insert.
▸ Day 7: Inject prostaglandin or GnRH.
The treatment duration with the progesterone insert is 5–8 days. Animals exhibiting estrus should be artificially inseminated.
Adverse Reactions
No adverse reactions have been observed at the recommended dosage.
Precautions
▸ Not suitable for cattle with vaginal malformations.
▸ Wear rubber gloves when handling this product.
▸ Animals in poor health (eg, disease or nutritional deficiency) may not respond.
▸ Keep out of reach of children.
